Is it possible to become a Drill Instructor in the Guard?

Yes it is, but first you have to become a NCO and then you can goto Drill Sergeant School.

So if I enlist as prior service E5 is it possible to request DI school after WTC and AIT?

With few exceptions, to have a shot at DS School you will need to be assigned to a unit that has them slotted, such as a training unit. In some cases the state RTI's (Regional Training Institutes) may have a slot available for you to pursue - this is one where you have to talk to your recruiter about the units in your state and the MOS requirements of those units.
Your recruiter can help you to locate any slots that may exist that have a requirement for DS qualification. If there are none, then the chances of you getting the school and qualification are slim (if at all).
